In clinical trials, once-weekly semaglutide with lifestyle changes led to an average weight loss of around 15% at 68 weeks. For adults with excess weight and established heart disease, semaglutide also lowered major cardiovascular events by about 20%.
Most side effects are temporary stomach symptoms, nausea, vomiting, diarrhoea, often eased by slow dose increases. Semaglutide isn’t for everyone: avoid if you or a family member has medullary thyroid cancer or MEN2; use caution with a history of pancreatitis or gallbladder disease; and it isn’t recommended during pregnancy. Current FDA review has not found evidence that GLP-1 drugs cause suicidal thoughts, but patients should report new mood changes.
